表油菜素內酯(EBR)是油菜素類固醇,類固醇衍生植物生長調節劑家族的生物活性化合物。將細胞與各種劑量(0-100μM)的表油菜素內酯一起溫育24或48小時,并通過MTT測定法測定細胞活力。與LNCaP和DU145前列腺癌細胞中的未處理樣品相比,表油菜素內酯以劑量和時間依賴性方式誘導細胞活力喪失。與DU145細胞相比,增加濃度的表油菜素內酯對LNCaP細胞活力喪失更有效,這表明雄**依賴性細胞對表油菜素內酯比對雄**非依賴性前列腺癌細胞更敏感。在進一步的實驗中,選擇25μM的表油菜素內酯,因為其對兩種細胞系具有中等的細胞毒性作用。通過在96小時內計數細胞數來檢查表油菜素內酯治-療對細胞增殖的影響。在LNCaP中觀察到比DU145細胞更高和更早的細胞增殖抑制[1]。
